Abstract
서론:
테모졸로마이드 치료 저항성은 다양한 DNA 복구 기전과 관련되어 있다. 본 연구에서는 테모졸로마이드 치료 저항성 획득 기전 중, 특히 상동재조합 DNA 복구 기전에 초점을 맞추어, HOX 유전자의 역할에 대해서 분석하였다. 또한 MGMT 유전자 프로모터의 메틸화 여부에 따른 HOX 유전자의 임상적 의미에 대해 연구하였다.

방법:
서로 다른 MGMT 유전자 프로모터의 메틸화를 가진 2개의 교모세포종 세포주를 이용하여, HOXA10 유전자의 억제에 따른 테모졸로마이드 항암요법의 치료 저항성의 차이에 대하여 연구하였다. 또한 교모세포종 세포주 실험을 통하여 RNA 간섭에 따른 유전자 발현의 차이를 분석하여 치료 저항성 획득 기전에 관한 연구를 진행하였다.

결과:
HOXA10 유전자의 억제는 각각의 세포주에 있어서 MGMT 프로모터 메틸화 여부에 상관없이 테모졸로마이드의 치료 감수성을 증가시키는 결과를 얻었으며, 이러한 치료감수성의 증가 기전은 Rad51 상동체의 하향조절로부터 얻어지는 DNA 손상 복구 과정의 결손과 관련이 있는 것으로 연구되었다. 특히, EGR1과 PTEN 유전자는 HOXA10 유전자와 상동재조합 DNA 복구기전 사이를 조절하는 중요한 매개 역할을 하고 있음을 확인할 수 있었다. 또한 HOXA10 유전자의 발현 억제는, 세포질내에서 이뤄지는 phosphoinositide 3-kinase / Akt 경로와는 무관하게 PTEN 유전자의 핵내 발현에만 선택적으로 작용하는 것으로 연구되었다.

결론:
본 연구에서 분석된 HOXA10 유전자에 의한 상동재조합 DNA 복구기전의 조절은 교모세포종 환자에 있어서 테모졸로마이드의 치료 저항성 극복을 위한 새로운 치료 전략이 될 수 있음을 보여주는 것이라 할 수 있겠다.
Introduction
Temozolomide resistance is associated with multiple DNA repair pathways. We investigated homeobox (HOX) genes for their role in temozolomide resistance, focusing on the homologous recombination (HR) pathway, and we tested their therapeutic implications in conjunction with O6-methylguanine DNA methyltransferase (MGMT) status.

Methods
Two glioblastoma cell lines with different MGMT statuses were used to test the augmented anticancer effect of temozolomide with HOXA10 inhibition. In vitro experiments, including gene expression studies with RNA interference, were performed to verify the related pathway dynamics.

Results
HOXA10 inhibition reinforced temozolomide sensitivity independent of MGMT status and was related to the impaired double-strand DNA breakage repair process resulting from the downregulation of Rad51 paralogs. Early growth response 1 (EGR1) and phosphatase and tensin homolog (PTEN) were the regulatory mediators between HOXA10 and the HR pathway. Moreover, HOXA10 inhibition selectively affected the nuclear function of PTEN without interfering with its cytoplasmic function of suppressing the phosphoinositide 3-kinase/Akt pathway.

Conclusions
The mechanism of HR pathway regulation by HOXA10 harbors another target mechanism for overcoming temozolomide resistance in glioblastoma patients.
