Abstract
본 연구에서는 국내·외 품종별 green pepper에 있는 비타민 C, 총 폴리페놀, 총 플라보노이드를 비롯하여 고추에 주로 함유된 flavonoids인 quercetin, luteolin, apigenin의 양을 측정하고, 이들로 인한 항산화 효과와 더불어 유방암 세포에서의 apoptosis를 통한 세포 사멸 유도에 관한 연구를 수행하였다. 이에 국내산 green pepper의 flavonoids에 대한 기초 자료를 제공하고, 이들의 항산화 효과 및 유방암 세포에서의 apoptosis를 통하여 암을 비롯한 다양한 질병의 예방에 있어 잠재적인 가치를 알아보고자 하였다.
풋고추, 청양고추, 꽈리고추, 오이고추, 할라피뇨의 5가지 품종의 flavonoids를 분석한 결과, 그 함량에 있어서 luteolin, quercetin, apigenin의 순으로 측정되었으며, 품종 간에는 꽈리고추의 flavonoids가 유의적으로 가장 높게 나타났다. 비타민 C의 함량은 꽈리고추가 가장 높았으며, 그 다음 청양고추, 할라피뇨, 풋고추, 오이고추의 순으로 측정되었다. 또한 총 폴리페놀과 총 플라보노이드 함량은 꽈리고추가 유의적으로 높게 측정되었는데 이는 DPPH, ABTS 자유기 소거 활성능에서도 비슷한 경향을 나타내었다.
Green pepper 추출물의 세포 성장에 미치는 영향을 살펴본 결과, human breast cancer cell인 MCF-7과 MDA-MB-231에서 5가지 품종 모두 농도 의존적으로 유의적인 증식 억제능을 갖고 있었다. 특히 꽈리고추에서 IC50 값이 826 μg/mL, 719 μg/mL로 가장 낮게 측정되어 증식 억제 효과가 가장 우수하게 나타났다.
Necrosis로 인한 세포 사멸 효과는 꽈리고추 추출물을 처리하였을 때, 농도 의존적으로 LDH가 분비되며, plasma membrane integrity의 감소가 이루어지는 것을 확인하였다.
Apoptosis의 확인 인자로 Bax, cleaved caspase-3, PARP를 측정한 결과, 꽈리고추 추출물에서 농도 의존적으로 apoptosis로 인한 세포사멸 효과를 나타내었다.
이상의 결과들을 통하여, green pepper에는 luteolin, quercetin 및 apigenin의 flavonoids를 비롯하여 vitamin C 등 여러 기능성 성분을 함유하고 있으며, 특히 꽈리고추가 5가지 품종 가운데 항산화 및 항암능 측정에서 가장 높은 활성을 갖고 있었으며, 유방암세포에서 apoptosis로 인한 세포 사멸 효과를 나타내고 있는 것을 확인하였다.
이에 국내산 green pepper의 flavonoids의 정성·정량에 있어서 기초자료가 될 것으로 기대되며, 이들의 항산화 효과와 더불어 유방암 예방 및 치료에 있어서 잠재적인 가치를 기대할 수 있다.
Pepper (Capsicum annuum. var.) has various phytochemicals including capsaicin and flavonoid. However, studies on flavonoids in pepper are a few, compared to many researches done on capsaicin. So, we investigated total phenol, total flavonoid contents, antioxidant activity and antiproliferative activity though apoptosis on human breast cancer cell line (MCF-7, MDA-MB-231). This study was carried out to determine whether green pepper have a potential to prevent proliferation of human breast cancer.
Four varieties of Korean green peppers (PP : Phut Pepper, CP : Cheongyang pepper, KP : Kkuri Pepper, OP : Ohi pepper) and one foreign green pepper (JP : Jalapeno Pepper) were used. Major flavonoids of green pepper were luteolin, quercetin, and apigenin. Content of luteolin was the highest in green pepper, especially in KP. The amounts of vitamin C were the highest in KP, followed by CP, JP, PP, and OP. The contents of total phenol were the highest in KP. KP had highest contents of total flavonoid. For antioxidant activity using DPPH·ABTS radical scavenging activity, KP had the most potent antioxidant activity, indicating the correlation between the content of flavonoid and vitamin C and the antioxidant activity.
In the result of antiproliferative activity on human breast cancer cell, KP had the highest antiproliferative effect (IC50 826.07 μg/ml in MCF-7, 719 μg/mL in MDA-MB-231).
KP extracts exhibited moderate cytotoxicity caused by necrosis against the cancer cell lines, in a dose-dependent manner, as measured by against the untreated control.
To confirm the apoptotic activity, modulations of Bax, cleaved caspase-3, and PARP enzyme expression were evaluated by western blot, and KP showed dose dependent induction of apoptosis on breast cancer cell.
These results suggest that green pepper has many functional compounds including flavonoids such as luteolin, quercetin, and apigenin, and vitamin C. Also, KP had the highest antioxidant activity and apoptotic activity among the five tested varieties.
In conclusion, green pepper has significant antioxidant activity and can be a possible candidate for treatment of breast cancer.
