dc.description.abstract조직공학에 있어서 세포의 부착과 증식, 분화를 위하여 골의 세포외기질과 유사하게 형성된 3차원적 지지체의 필요성이 높아지고 있다. 콜라겐 지지체는 매우 다공성인 특성을 지녀 조직공학용 지지체로 가장 많이 쓰이고 있는데, 콜라겐 지지체의 성능을 향상시키기 위해 생리활성 펩타이드의 활용이 필요한 상황이다. 본 연구에서는 mineralized connective tissue에 특히 많이 분포하여 골 경화의 결정핵 형성을 촉진하는 Bone sialoprotein 구조 중에서 기능성 펩타이드를 도출하고 합성하여 그 기능을 확인하고자 하였다. 또한 생리활성 펩타이드가 고정된 콜라겐 지지체의 골조직재생능을 관찰하여 향후 현재 동종이식재나 자가이식재가 필요한 골대체 및 재생영역에 있어서 활용 가능성을 고찰해보고자 한다.

콜라겐 지지체를 제조하고, 펩타이드를 합성하기 위해 Bone sialoprotein(BSP)의 구조 중 경조직형성능 및 콜라겐에 부착하는 부위에 해당하는 아미노산 서열을 도출한 후, 이를 펩타이드 합성기로 대량 합성하였다. 콜라겐 지지체에 얻어진 펩타이드를 고정한 후 human Mesenchymal Stem Cell(hMSC)을 분주하고 세포부착을 관찰하였다. 세포부착을 관찰하기 위하여 전자현미경(SEM)을 사용하였다. MTT assay를 통하여 세포독성 및 부착능을 측정하고 생리활성 펩타이드 자체의 골조직재생능을 평가하기 위하여 웨스텐블랏법(Western Blotting Assay)를 시행하였다. 마지막으로 콜라겐 지지체에 펩타이드를 고정하여 hMSC를 분주, 부착시키고 Calcein Staining 후 Confocal Microscopy를 이용하여 BSP에서 분리된 생리활성 펩타이드가 골조직재생능에 미치는 영향을 관찰하였다.

BSP로부터 유래된 합성펩타이드는 그 자체로 세포독성을 일으키지 않았고 콜라겐 지지체에 안정적으로 고정되었다. hMSCs를 분주하였을 때 대조군인 합성펩타이드 미처리 지지체에 비하여 세포 부착이 현저히 증가되었고 골조직재생능 또한 향상되었다. 따라서 BSP로부터 유래된 합성펩타이드는 콜라겐 지지체에 안정적으로 고정되며 골조직재생능도 향상시켜 향후 in vivo 평가가 이어진다면 골재생영역에서 활용가능성이 높을 것이라 생각된다.

dc.description.abstractThere is a need in tissue-engineering for 3D scaffold that recreate the natural extracellular matrix of bone to promote cell adhesion, proliferation, and differentiation. Collagen is highly porous, but its mechanical properties is very weak. To enhance the bioactivity of scaffolds, we investigated a synthetic oligopeptides derived from bone sialoprotein. Bone sialoprotein is expressed at high levels in mineralized connective tissues, promotes mineralization and nucleatrion. The aim of this study was to evaluate the osteogenic capacity of collagen scaffolds coated with collagen binding peptide in vitro study.

A peptide sequence corresponding to the collagen-binding domain of bone silaoprotein, was synthesized chemically using an automatic peptide synthesizer. Human mesenchymal stem cells were used to evaluate the cell adhesion, proliferation and differentiation activity of the peptide-coated collagen scaffolds. The cell adhesion was observed by SEM. MTT assay demonstrated that peptide supported cell adhesion and viability. The osteogenic capacity of peptide-coated collagen scaffolds was evaluated by using Alizarin S red staining, ALP staining and confocal microscopy.

The synthetic peptide corresponding to collagen binding domain from BSP was stably immobilized to the collagen scaffolds. The cell adhesion and differentiation of peptide-coated collagen scaffolds were slightly enhanced than that of collagen scaffolds without peptide. Therefore, the synthetic peptide would be used as a new biomaterial in the future.
