Studies on electrophysiological properties of Dscam / Vps13b mutant mice

Abstract

자폐 스펙트럼 장애는 사회적 상호작용, 인지기능에 손상이 있거나 반복적인 행동을 보이는 것을 특징으로 한다. 최근 여러 유전자 분석 연구를 통해 자폐 스펙트럼 장애를 가진 환자들에서 VPS13B 유전자의 변이와 DSCAM 유전자의 변이가 발견되었다는 보고가 있다. 본 연구에서는 복합적인 시간적, 비용적, 윤리적 문제로 인해 인간 모델이 아닌, 인간과 유사한 특징을 가진 마우스 모델을 사용하여 두 유전자의 전기생리학적인 특성을 살펴보았다. 이를 통해 Vps13b 유전자 적중 마우스에서는 야생형 마우스와 비교하였을 때 미세 흥분성 시냅스 후 전류의 진동수가 증가하여 시냅스 전 뉴런에서 신경전달물질의 분비가 증가하였을 것임을 확인하였다. Dscam 유전자 적중 마우스의 경우, Vps13b 유전자 적중 마우스와 다르게 미세 흥분성 시냅스 후 전류의 진동수와 진폭에서 유전자형 간에 큰 차이는 없었다. 하지만, 기존의 자폐 스펙트럼 질병 연구들에서와 동일하게 NMDA/AMPA 비율이 감소한 것을 확인하였다. Dscam 유전자 적중 마우스의 경우 NMDA 수용체 구성요소의 단백질 양에서는 유의미한 차이가 없었으나, 뇌의 신호 전달 과정에서 확연한 변화가 있음을 볼 수 있었다. 따라서, Dscam 유전자 적중 마우스가 자폐 스펙트럼 장애의 마우스 질병 모델로서 사용 가능성을 가짐을 보여주었다.

Features of Autism Spectrum Disorder (ASD) include deficits in social communication skills, impaired cognition, and repetitive behavior. Recent findings reported that ASD patients have mutations in VPS13B gene or DSCAM gene. In this study, two mouse models were used to identify electrophysiological changes because mice share similar genetic characteristics with humans. The frequency of miniature excitatory postsynaptic currents (mEPSC) was increased in Vps13b KO mice, entailing an increase in the neurotransmitter release. However, there were no differences in basal synaptic transmission in Dscam KO mice. Instead, the NMDA/AMPA ratio was significantly decreased in Dscam KO mice. These results are consistent with previous research on ASD. The DSCAM gene strongly effected electrophysiological properties although there were no changes in protein level of NMDA receptor components. Therefore, Dscam KO mice have potential as an ASD mouse model.
