Abstract
The movement of various muscles, such as grasping or walking, is accomplished via a well-organized system where the axons of motor neurons from central nervous system (CNS) stimulate various target muscles. Each motor neuron acquires diversity through the combination of various transcription factors during embryo development. Sonic hedgehog (Shh) secreted from the floor plate at the early developmental stage of the spinal cord generates a concentration gradient along the dorsoventral axis of the spinal cord and contributes to the progenitor domain formation of the ventral neural tube.
During this process, motor neurons are derived from the motor neuron progenitor (pMN). The basic helix-loop-helix (bHLH) transcription factor, Olig2, which is expressed in pMN, induces the expression of ISL1 and LHX3 in the final mitosis of pMN. Comprised of these two factors, the ISL1-LHX3 complex recognizes the hexamer response element (HxRE) of the genes related to giving identities to the motor neurons and is involved in the transcriptional regulation of these genes. Among number of target gene candidates, this study investigates the role of STAM1 and ARHGAP36 in motor neuron specification and maturation.
Studies have shown that STAM1 participates in endocytosis and, together with HRS, forms an endosomal sorting complexes required for transport (ESCRT-0) complex, which is essential in generating multivesicular bodies and cytokinesis. This research reveals that the ISL1-LHX3 complex binds to the enhancer of STAM1 and directly regulates the expression of STAM1, specifically in the motor neurons of the spinal cord. CXCR4, which is recognized by ESCRT-0, is known to be important for axons to reach the ventral exit point of the spinal cord and project their branches to the targeted muscle during motor neuron development. In the spinal cord motor neurons of chick embryo, CXCR4, a target protein of ESCRT-0, is also expressed along with STAM1 and HRS, and there exists an interaction between them. When STAM1 is specifically knocked down in the chick spinal cord, CXCR4 protein level shows positive correlation with STAM1. This results in motor neurons to fail in finding the ventral exit point and project their axons to dorsal spinal cord. Similarly, overexpression of STAM1 or CXCR4 shows similar phenotypes. Based on the above results, this study shows that STAM1, the one of the target genes of the ISL1-LHX3 complex, is vital in maintaining the proper amount of CXCR4 and leads motor neurons to exit the spinal cord through the correct exit point.
Another target gene, ARHGAP36, also shows that ISL1-LHX3 binds to the HxRE present in the enhancer region to regulate the transcription of the gene. ARHGAP36 is expressed only in the motor neurons in the spinal cord and more specifically, in the lateral motor column (LMC). Previous studies have shown that ARHGAP36 is known to mediate Shh signaling as a downstream factor of the Shh pathway, but the importance of ARHGAP36 and Shh signaling in postmitotic motor neurons is not well known. The Shh or Arhgap36 knockout mouse model failed to maintain the normal levels of LMC and overexpression of ARHGAP36 in chick embryo spinal cord showed an increase in LMC motor nerve. Also, AKT, a positive regulator of the Shh pathway, increases ARHGAP36 protein stability by inducing phosphorylation of ARHGAP36. As consequence, ARHGAP36 mediates Shh signal by inhibiting the kinase activity of PKA, a negative regulator in Shh pathway. This study shows that Shh-AKT signaling in LMC is capable of increasing the protein stability of ARHGAP36, whose expression is regulated by ISL1-LHX3 complex, and is an essential factor in maintaining LMC motor neurons.
In summary, STAM1 and ARHGAP36, which are the target genes of the ISL1-LHX3 complex, contribute to the specification and maturation of motor neurons, thereby indicating that they are important factors that give common characteristics to the motor neurons while generating diversity in each of them.
손을 움켜 쥐거나 걷기와 같은 다양한 근육의 움직임은 뇌와 척수로 구성되는 중추 신경계 (Central Nervous System)에서 운동 신경이 axon을 돌출하여 표적 하는 여러 근육들을 자극 시키는 조직적인 체계를 통해 이루어진다. 각 운동 신경은 embryo의 발달 과정 중에 다양한 전사 인자들의 조합으로 다양성을 얻게 된다. spinal cord의 초기 발달 단계의 floor plate에서 분비 되는 Sonic hedgehog (Shh)는 spinal cord의 dorsoventral axis를 따라 농도 구배를 이루며 ventral neural tube의 progenitor domain 형성에 기여한다. 이때 운동 신경 세포는 운동 신경의 progenitor (pMN)에서 유래하게 된다. pMN에서 발현되는 basic helix-loop-helix (bHLH) 전사 인자, Olig2는 pMN의 최종 세포 분열 과정에서 ISL1과 LHX3의 발현을 유도하게 되고 이 두 단백질이 모여서 만들어진 ISL1-LHX3 complex는 운동 신경에 정체성을 부여하는 관련된 유전자의 hexamer response element (HxRE)를 인식하여 해당 유전자의 전사 조절에 관여한다. 본 연구는 다양한 유전자 후보들 중, STAM1과 ARHGAP36가 운동 신경의 분화와 성숙에 어떻게 기여하는지 그 기능을 알아보고자 했다.
STAM1은 세포내이입에 관여하는 단백질로서 HRS와 함께 endosomal sorting complexes required for transport (ESCRT-0) 복합체를 형성, 다소포체 생성 경로와 세포질 분열에서 중요한 역할을 한다고 알려져 있다. STAM1의 enhancer에 ISL1-LHX3 complex가 결합하여 전사가 조절되고 spinal cord 내 운동 신경에서 특이적으로 발현됨을 밝혔다. ESCRT-0에 의해 인식되는 CXCR4는 운동 신경의 발달 과정 중 axon이 spinal cord의 ventral exit point를 찾아 나감으로써, 표적 하는 근육에 도달하는데 중요하다고 알려져 있다. Chick embryo의 spinal cord 운동 신경에서 STAM1, HRS와 함께 ESCRT-0의 표적 단백질인 CXCR4도 발현되고 있음과, 이들 사이의 상호작용이 존재함을 보였다. Chick embryo의 spinal cord에서 STAM1을 knockdown 시켰을 때, CXCR4의 단백질 양에 영향을 주고, 그로 인해 ventral exit point를 찾지 못한 운동 신경의 axon이 dorsal 방향으로 뻗어가는 현상을 관찰할 수 있었다. 마찬가지로 STAM1 혹은 CXCR4를 과발현 시켰을 때도 이런 유사한 phenotype을 보였다. 본 연구는 위의 결과들을 토대로 ISL1-LHX3 complex의 표적 유전자인 STAM1은 적절한 CXCR4의 양을 유지시켜 운동 신경이 올바른 exit point를 통해 spinal cord 밖으로 나갈 수 있게 유도함을 밝혔다.
또 다른 표적 유전자인 ARHGAP36도 enhancer region에 존재하는 hexamer response element (HxRE)에 ISL1-LHX3가 결합하여 Arhgap36의 전사를 조절함을 보였다. ARHGAP36는 spinal cord 내 운동 신경에서만 제한적으로 발현되고, 특히 Lateral Motor Column (LMC)에 특이적으로 발현하고 있음을 확인했다. 기존의 연구 결과에 따르면 ARHGAP36는 Shh pathway의 하위 인자로 Shh 신호 전달을 매개하는 역할을 한다고 알려져 있으나 postmitotic 운동 신경에서 ARHGAP36와 Shh 신호의 중요성은 잘 알려지지 않았다. Shh 혹은 Arhgap36 knockout mouse model은 정상적인 수준의 LMC를 유지하지 못하고 chick embryo spinal cord에서 ARHGAP36의 과발현은 LMC 운동 신경의 증가를 유도함을 보였다. 또한 Shh pathway의 positive regulator인 AKT가 ARHGAP36의 인산화를 유도함으로써 protein stability를 증가시키고 결과적으로 ARHGAP36는 PKA의 kinase activity를 억제시켜 Shh 활성을 매개함을 발견했다. 본 연구는 ISL1-LHX3 complex에 의해 발현이 조절된 ARHGAP36가 LMC에 존재하는 Shh-AKT 신호를 통해 protein stability가 증가되고 LMC 운동 신경이 유지되는데 필요한 요소임을 밝혔다.
종합하면, ISL1-LHX3 complex의 표적 유전자들인 STAM1과 ARHGAP36는 운동 신경의 분화와 성숙에 기여함으로써 운동 신경들이 갖는 공통적인 특성을 부여함과 동시에 각 운동 신경만의 다양성을 생성하는데 중요하다는 것을 보여준다.
