Abstract
Cystic disease of the kidney was induced in the
newborn white rabbits by a single intramuscular inj·
ection of prednisolone crystalline (50mg/kg) , cortisone
acetate (200mg/kg) , dexamethasone (40mg/kg) ,
methylprednisolone acetate (20mg/kg) , and their conventional
light microscopic and ultrastructural changes
were constructed to understand the morphogenesis
and the sequential evolution of cystic lesions
1. Cystic lesions were induced only in the perinatal rabbits by injection of methylprednisolone acetate
(MPA) and dexamethasone, and not by prednisolone
crystalline or cortisone acetate. All pregnant animals
with MPA injection resulted in total failure by abor'
tion.
2. Cystic lesions were predominantly located in the
renal cortex, and consisted of 2 major types; the
tubular cyst of proximal and collecting tubular origin
in the early phase, and the glomerular cyst of diIat·
ation of Bowmans urinary space in the latcr phase.
3. Tubular dilatation was most profound at the 3rd
week after injection and showed a tendency of reduction
thereafter. On the contrary, the glomerular cysts
developed with increasing tendency from the 4th week.
4. Cystic dilatation of the proximal and collecting
tubules was more prominent in rabbits with prolongation
of perinatal period of nephrogenic activity in
the renal cortex; there were also morphological evid
ences of interlesional continuity between the nephrogenic
vesicle or S-shaped bodies and cystic structures.
5. Ultrastructurally poor development ef microvilli
of proximal tubules, numerical decrease and irregula·
rity of intracytoplasmic organelles, and delayed development
of baso-Iateral infoldings were all indicative
of maturation arrest, and also were associated with
uneven thickening of tubular basement membrane
together with lamellation, fraying and rarefaction
With the above findings it is the author's assumption
that steroid hormone, especially MPA may result in
maturation arrest of nephrogenic activity in the perinataI
renal cortex, and its cystic evolution is closely
correlated with alteration of tublar basement membrane
during the period of nephrogenic activity.
