Thrombospondin-1 prevents Aβ-mediated synaptic pathology in Alzheimers disease : 알츠하이머병에서 아밀로이드 베타 단백질에 의한 시냅스 독성 작용에 있어서 thrombospondin-1의 방어기전 연구

- Abstract
- 알츠하이머병은 인지능력 장애와 기억소실 등으로 특징 지어지는데, 시냅스의 병리학적 현상에 의해 일어나는 것으로 알려져 있다. Thrombospondin은 성상 교세포에서 주로 분비되는 물질로서 혈관 형성을 억제를 일으키는 기능이 있는 것으로 잘 알려져 있는데, 최신 보고들은 혈관형성에 관련된 역할이 아닌 시냅스의 형성과, 신경세포의 발생 등이 다수이다. 본 연구는 thrombospondin의 다섯 가지 isoform 중 하나인 TSP-1의 역할에 대해 in vitro 와 알츠하이머 동물모델을 이용하여 진행하였다. Astrocyte에서 TSP-1의 분비가 아밀로이드 베타 단백질에 의해 감소하는 것을 in vitro 상에서 관찰 하였고, 알츠하이머병 동물모델인 5xFAD mice와 Tg2576 mice 에서 TSP-1의 양이 감소되어 있는 것을 관찰 하였다. 또한, 시냅스의 병리학적 현상인 수상돌기의 밀도 감소, 시냅스에서의 소포체 재순환의 감소, 그리고 LTP 등이 아밀로이드 베타 단백질에 의해 감소하는 것을 규명하였고, TSP-1과의 공동 배양에 의해 아밀로이드 베타 단백질의 영향으로부터 보호되는 것을 규명 하였다. 결론적으로, TSP-1이 아밀로이드 베타 단백질에 의해서 일어나는 신경세포 및 시냅스의 손상을 보호 함으로써, TSP-1의 생성 및 분비 감소 등이 알츠하이머병의 병인기전에 밀접하게 연관됨을 규명하였다.
Alzheimers disease (AD) is characterized by cognitive dysfunction and memory loss, which are often the result of synaptic pathology. Thrombospondin (TSP) is an astrocyte-secreted protein well-known for its function as an anti-angiogenic role. Recently, however, the role of TSP as a modulator of synaptogenesis and neurogenesis has been highlighted. Here, I studied the role of one of the five TSP isoforms, TSP-1, in vitro and in AD animal models. TSP-1 release was decreased by Aβ42 in vitro and the reduced level of TSP-1 was observed in both 5xFAD and Tg2576 mice. Synaptic pathology caused by Aβ42 such as decreased dendritic density, impaired vesicle recycling, and reduced long-term potentiation (LTP) were prevented by co-incubation with TSP-1 and Aβ. TSP-1 is a potential therapeutic component against the damaging effects caused by Aβ42 in AD pathogenesis.
