dc.description.abstract망개나무 (Berchemia berchemiifolia (Makino) Koidz.)는 갈매나무과 (Rhamnaceae) 망개나무속 (Berchemia)의 국내에서 자생하는 낙엽교목이다. 2005년에는 멸종위기 야생동·식물 Ⅱ급으로 지정되었을 만큼, 그 수가 많지 않았지만, 그 수가 안정되어 2012년에 해제되었다. 망개나무속에서는 anthraquinone, ventiloquinone 등의 quinone 계열 화합물과, flavonoid 등의 phenolic compound가 분리, 보고되었으며, 망개나무속 식물들은 예로부터 중국과 일본 등지에서 복통, 관절염 등의 치료에 사용되었다고 한다. 그러나, 망개나무속의 망개나무에 대한 연구는 거의 이뤄지지 않은 상태로, 본 연구에서는 한국 자생식물인 망개나무의 성분을 연구하고자 하였다.
망개나무 지하부의 80% 메탄올 추출물을 n-Hexane, Methylene chloride (CH2Cl2), n-BuOH로 분획하였었으며, 각각의 Mass spectroscopy 데이터를 통해 특이적인quinone과 secoiridoid 피크를 확인하여 이들을 타겟으로 분리를 진행하였다. 그 결과, CH2Cl2 층에서 총 3종의 quinone heterodimer, ventiloquinone-dihydroanthracenone dimer 화합물 (1) 과 ventiloquinone-naphthoquinone dimer 화합물 (2, 3), 3종의 anthraquinone 계열 화합물 (4-6), 그리고 1종의 flavonoid 계열 화합물 (14)을 분리하였으며, n-BuOH 층에서는 7종의 secoiridoid glycoside 계열 화합물 (7-13)을 분리하였다.
각 화합물은 berfoliquinone (1), berchemiquinone A (2), berchemiquinone B (3), aloesaponarin I (4), 3,8-dihydroxy-6-methoxy-1-methylanthra-9,10-quinone-2-carboxylic acid methyl ester (5), 4,8-dihydroxy-3-methoxy-1-methylanthra-9,10-quinone-2-carboxylic acid methyl ester (6), berfolioside A (7), berfolioside B (8), dihydroxyframoside A (9), insularoside (10), framoside (11), hydroxyframoside A (12), hydroxyframoside B (13), naringenin (14) 으로 동정하였다. 이들 중 화합물 1-3, 7-9는 천연에서 처음으로 분리, 보고되는 물질이다. 특히 화합물 2와 3은 천연에서 처음 분리 보고되는 3원환 - 2원환 quinone heterodimer 이다.
분리한 quinone 계열 화합물 (1-5) 을 대상으로 HCT-116 인간 대장암 세포에 대한 세포독성 활성을 측정하였으며, 이들 중 2와 4가 비교적 유의미한 활성을 나타내었다. Secoiridoid glycoside 계열 화합물 (7, 8, 10, 11)을 대상으로는 LPS 유도된 RAW 264.7 마우스 대식세포에 대해 NO 생성 억제을 측정하였으며, 그 중 11이 비교적 NO 생성을 억제하는 것으로 나타났다.

dc.description.abstractBerchemia berchemiifolia (Makino) Koidz. is a deciduous tree of the genus Berchemia which belongs to the Rhamnaceae family. Because of its small population, it was designated as endangered species in 2005, but was removed in 2012 as the population stabilized. In the previous isolation studies of genus Berchemia, quinone-related compounds such as anthraquinones and ventiloquinones, phenolic compound such as flavonoids were reported. The plants in the genus Berchemia have been used in China and Japan to treat abdominal pain and arthritis. However, little research has been conducted on B. berchemiifolia.
In this research, fourteen compounds, six quinones (1-6), seven secoiridoid glycosides (7-13) and one flavonoid (14) were isolated from the roots of B. berchemiifolia. Among them, berfoliquinone (1), berchemiquinone A (2), berchemiquinone B (3), berfolioside A (7), berfolioside B (8) and dihydroxyframoside A (9) were reported for the first time from nature. The quinones were tested for their cytotoxicity against HCT-116 human colorectal carcinoma cell lines, and for the secoiridoid glycosides, the anti-inflammatory activity were evaluated. Compounds 2 and 4 showed moderate cytotoxic activity against human colorectal carcinoma cell line HCT-116 and compounds 7 and 11 showed weak inhibition of NO production in RAW 264.7 cell line.
