Characterization of viral factor(s) required for systemic infection and different symptom development in Chenopodium quinoa by using : Tomato bushy stunt virus 와 Grapevine Algerian latent virus 의 흰 명아주에서 나타나는 전신감염과 병징 차이에 관여하는 바이러스 인자 연구

Abstract
Tomato bushy stunt virus (TBSV) and Grapevine Algerian latent virus (GALV) isolates, which belong to the the genus Tombusvirus, was collected in Korea. Infectious clones for both isolates, pTBSV-Sa and pGALV-Na, were generated and infectivity assay was carried out on various host to investigate the host range. Interestingly, these two isolates showed different symptom on Chenopodium quinoa. pTBSV-Sa induced local lesion while showing systemic infection by pGALV-Na. Deduced amino acid sequence alignment of capsid protein (CP) and p19 revealed a relatively low sequence identity of about 47% and 74%, respectively, while RdRp and movement protein showed high identity of about 97% and 83%, respectively. To determine whether these amino acid differences of CP and p19 between these two isolates involved in symptom development and/or systemic movement, deletion as well as gene replacement clones were generated and tested on C. quinoa. GALV CP deletion clone, pGMG (GALV CP deletion), did not move systemically suggesting that the possible role of the CP of GALV in systemic movement of the virus. A total of eight chimeric clones were generated by exchanging each domain of TBSV and GALV. We observed unexpected symptoms from plant inoculated by several chimeric clones suggesting the involvement of the other viral product(s) in symptom expression as well as systemic movement. Construction of an additional clones containing gene replacement between pGALV and pTBSV and inoculation experiments with generated clones revealed p19 as an additional determinant for regulating systemic invasion on C. quinoa.
Tombusvirus는 세계적으로 가장 잘 연구된 바이러스 중이 하나로, 토마토와 가지에 큰 피해를 끼친다. 최근 경상남도 사천에서 토마토에 피해를 주는 Tomato bushy stunt virus (TBSV)와 전라남도 나주에서 화훼작물에 피해를 주는 것으로 보고된 Grapevine Algerian latent virus (GALV)를 이용하여 먼저 지표식물들에 병원성 검정 실험을 실시 하였다. 검정 결과, 병원성의 약간 차이는 있었지만, 담배류의 식물에서는 유사한 병징을 나타내었다. 그러나 놀랍게도 Chenopodium quinoa (흰 명아주)에서는 TBSV-Sa는 괴사 반점을 동반한 국부병징을 GALV-Na는 상엽에서 황화반점을 동반한 전신병징을 나타내는 것을 관찰하였다. 이러한 결과를 토대로 먼저 바이러스의 어떤 인자가 이러한 병징 차이에 관여하는지 확인해 보기 위해서 두 바이러스의 아미노산 염기서열을 비교 분석해 본 결과, RNA-dependent RNA polymerase(RdRp), Capsid protein(CP), Movement protein(MP), p19은 각각 98%, 47%, 87%, 74%의 유사도를 보였으며, 일반적으로 Tombusvirus에서는 CP 혹/그리고 p19는 기주의 종에 따라 전신병징에 관여 된다고 보고 되어왔으나, 우선 가장 낮은 유사도를 보인 외피단백질이 이러한 병징의 차이를 나타내는 요소라 사료 되어, 이를 위해 감염성 클론 (pTBSV-Sa, pGALV-Na)이 제작되었고, 또한 외래 유전자가 식물에서 발현 할 수 있도록 외피단백질이 제거된 벡터(pTMT, pGMG)가 제작 되었고 흰 명아주에 감염 결과, pGMG(GALV 외피단백질 삭제)경우 단지 접종엽에서 병징이 야기되는 국부병징이 나타나 그 결과, GALV 외피단백질이 흰명아주의 전신병징에 관여하는 인자임을 확인할 수 있었다. 그리고 흰 명아주에서 나타나는 병징의 차이가 외피단백질의 차이라는 것을 증명하기 위해 감염성 클론의 각 도메인을 교환시킨 총 8개의 키메릭 바이러스 클론(pGGG, pGTG, pGGT, pGTT, pTGG, pTTG, pTGT, pTTT)을 만들어 식물에 감염시킨 결과, 몇몇의 클론에서는 기대하지 않은 병징이 나타났으며 이 말은 곧 외피단백질이 아닌 다른 도메인이 전신병징에 관여 함을 확인할 수 있었다. 그리고 전신병징을 나타낸 키메릭 바이러스 클론(pGGG, pGTG, pTGG, pTTG)는 공통적으로 GALV의 p19을 가지고 있었고, 이를 확인하기 위해서 site-directed mutageenesis라는 방법을 이용하여 p19이 비활성화된 클론(pGGG19m, pGTG19m, pTGG19m, pTTG19m)을 만들어 흰 명아주에 감염 결과, 모두 전신병징이 아닌 국부병징을 나타내었으며, GALV p19이 활성화 된 클론들에 비해서 병징이 급격히 약화되는 것을 확인할 수 있었다. 그 결과, GALV의 외피단백질과 p19이 흰 명아주의 전신병징에 기여하는 바이러스 인자임을 확인할 수 있었고, 흰 명아주에서의 병징의 차이는 외피단백질이 아닌 p19 차이에 의해 야기된다는 것을 확인할 수 있다.
