저선량 알파선에 대한 세포 반응의 선량률 효과 연구 : Study on Alpha Particles Dose Rate Effect of Cellular Response in Low Dose Exposure

Abstract
Alpha particles exposure mainly occurs as an internal exposure on human body. Mostly, the epidemiological studies and in vitro studies with very low dose range have been proceeded to find the biological responses of alpha particles exposure. However, the number of in vitro studies using alpha particles are relatively low compared to the in vitro studies using other types of radiation. Therefore, the in vitro studies about cellular responses of alpha particles are necessary. In this study, the in vitro experiments were proceeded to find the cellular response on the low dose of alpha particles and the effect of dose rates on the cellular response. Also, the effect of LET on the cellular response was observed.
In this study, rat diencephalon cell and rat gliosarcoma cell were used as a normal and a tumor cell. The SNU-ALPHACELL that is built in Radbio Labatory at Seoul National University was used as the alpha particle irradiator. Both cells were irradiated with the alpha particles at the dose rates of 0.05, 0.1, 0.5, and 1 Gy/min. 9 different doses, including a control group (0 Gy), were set for the cell irradiation. Survival were measured by clonogenic assay.
Both cells formed more colonies than the control group in the low dose (<0.2 Gy) range at the dose rate of 0.05 and 0.1 Gy/min and considered this phenomenon to be a Hormesis. The hormesis was not observed at the dose rate of 0.5 and 1 Gy/min and the cell survival curve followed a linear model. Through with this results, it was confirmed that as the dose rate increased, the hormesis disappeared. Also, comparing the slopes of the cell survival curves at the dose rate of 0.5 and 1 Gy/min, the slope at the dose rate of 0.5 Gy/min was steeper. The same trend was shown from the slopes of cell survival curve at 0.05 and 0.1 Gy/min. Therefore, the slope at 0.05 Gy/min was steeper.
It was assumed that hormesis was observed due to the low concentration of nitric oxide produced during low dose alpha particles exposure. The previous study found that the low concentration of nitric oxide increases the cell proliferation. Also, it was assumed that the slope difference of the cell survival curve for each dose rate was due to the LET (Linear Energy Transfer) difference between dose rates set as experimental conditions. So the RBE (Relative Biological Effectiveness) was calculated by measuring the LET values from each dose rate. The RBE increases exponentially for the LET and then decreases at the certain point. Since this experiment was performed in the section where the RBE decreases after the maximum value, the slope difference between the cell survival curves can be explained by the overkilling effect.
Although the public interest in the hazards of alpha particle exposure is increasing day by day, there are not enough researches on low dose alpha particles exposure. This is because the fact that the cell survival curve of high LET radiation exposure, such as alpha particles, follows the linear model was accepted without objection. However, the in vitro result in this study did not follow the existing model. The hormesis that was observed in this in vitro experiment should be considered in subsequent researches on the risk assessment of alpha particles exposure. Also, more accurate cellular responses can be identified by conducting more in vitro experiments with various dose conditions.
인체에 받는 피폭 중 알파선으로 인한 피폭은 주로 내부피폭에서 나타난다. 알파선 피폭으로 인한 생물학적 효과를 알아보기 위한 선행 연구로는 주로 역학조사와 극저선량 알파선 실험이 진행되어왔다. 저선량 알파선에 대한 in vitro 실험은 다른 종류의 방사선에 대한 실험에 비해 여전히 적으므로 알파선을 활용한 세포 반응 연구는 앞으로도 계속 진행되어야 할 필요가 있다. 본 연구에서는 저선량 알파선에 대한 세포 반응, 그리고 선량률의 영향을 알아보기 위한 in vitro 실험을 진행하였다. 또한, 알파선 조사 시에 LET(Linear Energy Transfer)에 변화를 주어 그에 따른 세포의 반응을 살펴보았다.
본 실험에서는 정상세포인 rat diencephalon cell과 종양세포인 rat gliosarcoma cell을 사용하였고, 서울대학교 방사선생명공학연구실의 알파선 조사 장치를 활용하여 다양한 선량률을 구현하여 실험을 진행하였다. 조사 조건으로 설정된 선량률은 0.05, 0.1, 0.5, 그리고 1 Gy/min이고 각각의 선량률에 따라 대조군을 포함하여 9가지의 다른 선량 조건으로 실험을 진행하였다. 조사된 세포들은 군집형성능 실험을 통해서 생존율을 확인하였다.
두 세포 모두 0.05와 0.1 Gy/min에 대한 저선량(< 0.2 Gy) 구간에서 대조군보다 더 많은 군집이 형성되었고 이를 호르메시스 효과라고 간주하였다. 0.5와 1 Gy/min의 저선량 구간에서는 호르메시스 효과가 관찰되지 않았고 세포 생존율 그래프의 형태가 선형 모델을 따랐다. 이에 대하여 선량률이 증가할수록 호르메시스 효과가 사라지는 것을 확인하였다. 또한, 0.5와 1 Gy/min에 대한 세포 생존율 그래프 기울기를 비교하였을 때, 0.5 Gy/min의 기울기가 더 가파르게 나타났다. 0.05와 0.1 Gy/min의 세포 생존율 그래프에서도 동일하게 0.05 Gy/min의 기울기가 더 가파르게 나타났다.
저선량 알파선에서 두 세포 모두 호르메시스 효과가 관찰된 이유는 저선량 알파선 피폭시 생성된 저농도의 Nitric Oxide로 인해서 세포 증식 능력이 증가하였기 때문이라고 추측하였다. 각각의 선량률에 대한 세포 생존율 그래프의 기울기 차이는 실험 조건으로 설정된 선량률 간의 LET의 차이로 인해 나타난 것으로 보고 LET에 대한 생물학적 효과비를 구하여 비교하였다. 생물학적 효과비는 LET에 대해 지수함수 형태로 증가한 뒤에 어느 지점 이후에는 다시 감소한다. 본 실험은 LET에 따른 생물학적 효과비가 최대값 이후 감소하는 구간에서 진행되었기 때문에 세포 생존율 그래프 간의 기울기 차이는 overkilling effect로 설명할 수 있다.
알파선 피폭의 인체 위해에 대한 대중의 관심은 날로 높아져가고 있음에도 저선량 알파선 피폭에 대한 연구가 많지 않다. 알파선과 같은 고LET 방사선 피폭시 세포 생존 곡선이 선형관계를 따를 것이라는 기존 모델이 이의 없이 받아들여졌기 때문이다. 본 연구는 기존 모델이 in vitro 실험결과와 일치하는지 확인하고자 진행되었고, 실험결과가 기존 모델과 다소 차이를 보이는 것을 확인하였다. 즉, 저선량 알파선 피폭시 세포 생존 곡선은 선형 관계를 따르지 않는 결과를 보였다. 이번 연구에서 확인된 저선량률 in vitro 실험에서 호르메시스가 관찰된 현상은 알파선 피폭시 위험도 평가에 관한 후속 연구에서 고려되어야 한다. 또한, 더 많은 조사 조건의 실험이 진행된다면 더 구체적인 반응을 확인할 수 있을 것으로 생각된다.
