dc.description.abstract본 연구에서는, γ-lactam-β-lactone bicyclic 골격을 가지는 cinnabramide 계열의 천연물들을 합성하기 위해 순차적인 3개의 탄소에 stereocenter를 도입하는 연구를 수행하였다. Cinnabaramide A와 그 유도체들을 20S proteasome의 저해제로 다발골수종 병에 대한 생물학적 활성이 있다고 알려져 있다. 알돌 기질을 만들고, C-2 stereocenter에 의한 선택적인 부분입체 aldol reaction 과 hydrolytic DKR를 통해 비대칭적인 pyrrolidinone 합성법을 개발하였다. 이 pyrrolidinone 공통 중간체로부터 t-butyl protection과 silyl group deprotection, DMP oxidation, barbier type allylation 으로 추가적인 2개의 stereocenter를 도입하고, 다음 t-butyl deprotection과정까지 총 5단계로 천연물인 cinnabaramide E를 12%의 전체 수율로 합성하였다. cinnabramide E로부터 lactonization을 통해 cinnabaramide A를 합성하였고, cinnbaramide A로부터 N-Acetyl-L-cysteine을 도입하여 cinnabaramide F를 합성하였다.-
dc.description.abstractIn this study, in order to synthesize natural products of the cinnabaramides having a γ-lactam-β-lactone bicyclic backbone, We introduce five stereocenters of cinnabaramides A, E, F. Derivatives are known to have biological activity as inhibitors of the 20S proteasome. First, We construct asymmetric pyrrolidinone core having 3 stereocenters. The key reaction is intramolecular aldol reaction of the C-2 stereodefined aldol substrate. And we introduce 2 stereocenters using diastereoselective allylation. Through this strategy, a 9-11 step total synthesis of cinnabaramide E, A, F were achieved with high selectivity from silyl-protected serine.-
