Telomere Elongation Induced by Reactive Oxygen Species in Malignant Progression of Hepatocellular Carcinoma : 활성산소에 의한 텔로미어 신장이 간에서의 악성 종양화에 미치는 영향에 대한 연구

dc.description.abstract | 악성종양(惡性腫瘍)의 대표적인 특징인 침습성(侵襲性)과 전이성(轉移性)은 암환자의 직접적인 사망 원인으로 알려져 있다. 전암(前癌) 상태에서 짧아진 텔로미어는 악성종양화의 과정에서 텔로머라아제에 의하여 그 길이가 회복된다. 그러나 텔로머라아제가 어떻게 활성화되는지에 대해서는 명확하게 알려진 바가 없다. 본 연구에서 사람의 간세포암 세포주 (肝細胞癌 細胞株)에서 활성산소 (活性酸素)가 TERT를 활성화시키고 그 결과 텔로미어가 길어지는 현상을 관찰하였다. 이 현상은 PI3K-Akt 신호 활성과 TERT 프로모터에서의 후성적 변화에 의하여 매개되었다. 그리고 누드 마우스 모델을 이용한 실험을 통하여, 활성산소에 의하여 신장된 (伸張) 텔로미어가 악성종양세포에서의 특징인, 세포 증식, 세포 이동, 세포 침습을 유도한다는 사실을 발견하였다. 이러한 동물실험으로부터의 결과뿐만 아니라 간암 (肝癌) 환자의 조직들로부터의 결과에서도 활성산소에 의해 신장된 텔로미어가 악성종양화를 유도한다는 현상을 발견하였다. 간암 환자의 조직에서 측정한 텔로미어 길이는 활성산소와 양의 상관관계가 유의하게 있고, 더 나아가 긴 텔로미어를 가지는 간암 환자들은 전체 생존률 (全體 生存率) 및 무재발 생존률 (無再發 生存率)이 통계적으로 유의하게 감소함을 발견하였다. 따라서 본 연구를 통해서, 활성산소를 억제할 수 있다면 텔로미어 신장이 방해되기 때문에 간암에서의 악성종양화를 효과적으로 억제할 수 있다는 것을 제시하였다. | - |
dc.description.abstract | The invasive and metastatic features of human malignant tumors are well-recognized and affect mortality in patients. During malignant tumor progression, activated telomerase leads to regaining the telomeres lost during the premalignant stage. However, it was unclear how telomerase becomes reactivated. I find that reactive oxygen species (ROS) upregulate telomerase reverse transcriptase (TERT), leading to telomere lengthening in human hepatocellular carcinoma (HCC) cell lines. These effects are accompanied by the PI3K-Akt signaling activation and the epigenetic changes on the TERT promoter. In nude mice model, lengthened telomeres by elevated ROS levels promote tumor cell proliferation, migration, and invasion, which are characteristics of malignancy. Moreover, the telomere lengths of liver cancer patients positively correlate with the ROS levels as well as the death and cancer recurrence rates. These results highlight the importance of telomere lengthening by ROS in malignant progression and suggest a therapeutic strategy against malignant liver tumor. | - |
