Abstract
본 연구에서는 자가 촉매작용으로 가수분해가 가능한 생체 환원성 유전자 전달 시스템을 개발하였다. 마이클 첨가 반응 (Michael addition reaction) 및 구아니딘화 반응(Guanidinylation)을 이용하여 합성된 poly(cystaminebisacrylamide-agmatine) (poly(CBA-AG)) 는 pDNA와 정전기적 상호작용을 통해 직경 200-300 nm의 양전하를 띄는 폴리플렉스를 형성할 수 있으며, 환원성 환경에서 구조 내 이황화 결합이 분해되면서 pDNA를 방출할 수 있는 것으로 나타났다. 또한 개질 전 고분자 poly(cystaminebisacrylamide-diaminobutane) (poly(CBA-DAB)와 달리 poly(CBA-AG)는 pH 7.4 PBS 상에서 즉각적으로 분해되는 거동을 보였는데, 이는 구조 내 구아니딘기의 자가 촉매작용에 의한 아마이드 결합의 가수분해의 결과임을 실험적으로 확인하였다. 고분자의 세포 독성은 세포에 따라 차이는 있으나 강한 세포 독성을 나타내는 PEI25K에 비해 미미한 수준이었으며, 그에 반해 transfection 효율은 poly(CBA-DAB)와 양성대조군으로 사용된 PEI25K 보다 높은 수준을 보였다. 그러한 원인으로써 구아니딘기에 의해 세포 투과성이 약간 향상된 것을 확인하였으나 그 정도 면에서 높은 transfection 효율의 타당성을 확보하기가 어려웠고, 이어서 세포 내 거동을 관찰한 결과 구아니딘기의 핵 국재화 능력을 통해 pDNA를 세포핵 내에 효율적으로 전달하는 것을 확인할 수 있었다. 더불어 세포 내 환원성 환경에서의 이황화 결합의 분해 작용과 구아니딘기에 의한 자가-촉매작용으로 인한 가수분해를 통해 폴리플렉스가 빠르게 해리되면서 이를 더욱 촉진하는 것으로 판단하였다. 또한 암세포를 선택적으로 사멸하는 아폽틴 (apoptin) 발현 유전자를 이용한 항암효과를 평가한 결과에서 poly(CBA-DAB)와 비해 압도적인 효율을 보였고, 이로써 poly(CBA-AG)의 유전자 전달 시스템으로서의 기대 효과를 확인할 수 있었다.
Self-destroying bioreducible polymer, poly(CBA-AG) was synthesized for gene delivery systems. It could form 200-300 nm sized and positively charged polyplexes with pDNA, which can release pDNA in reducing environment due to the internal disulfide bonds cleavage. poly(CBA-AG) showed spontaneous degradation profile in normal aqueous condition in contrast to backbone polymer, poly(CBA-DAB) lacking guanidine moieties. It is probably due to the self-catalyzing hydrolysis of internal amide bonds by guanidine moieties. The cytotoxicity of poly(CBA-AG) was examined to be cell-dependent but minimal unlike cytotoxic PEI25K. Transfection efficiency of poly(CBA-AG) was found to be highly enhanced in comparison with that of poly(CBA-DAB) and even higher than that of PEI25K. In addition, therapeutic effect of poly(CBA-AG) polyplexes was verified by using apoptosis assay methods in vitro. In futher experiments, it was proved that these results were derived from the improved cellular uptake efficiency of the polyplexes by the guanidine groups. Moreover, it was also observed by confocal microscopy that pDNA delivered by poly(CBA-AG) was strongly accumulated in cell nuclei in contrast to the results of PEI25K and poly(CBA-DAB) showing pDNA located mainly in cytosol. These results indicate that high transfection efficiency of poly(CBA-AG) may be derived from the enhanced cellular uptake efficienct and the efficient pDNA localization in cell nuclei by guanidine moieties. Furthermore, it could be facilitated by the polyplexes dissociation via self-catalyzing hydrolysis by guanidine moieties as well as disulfide bonds cleavage in cytosol.
