Abstract
To terminate the transcription of mRNA transcript, it is important that Rat1 and Rai1 make complex and show catalytic activity in Saccharomyces cerevisiae. At this point, Rtt103 recognizes the CTD of RNA polymerase II. Meanwhile the existence of Rtt103 rises up, their exact role is not studied. In this study, by classifying the Rtt103 domain, we try to find their interacting partner and relate their role in transcription process. Elucidated proteins are one of capping enzyme in early stage of transcription, Ceg1 and other transcription factors. Understanding the relation of Ceg1 and Rtt103 provides a insight of whole process of transcription. The other is role of Rtt103 in Rat1/Rai1 complex for transcription termination. Confirming the Rtt103 surface with Rat1 complex by yeast hybrid system and in vivo. Rat1 and Rtt103 interaction is mediated by Rtt103 SCOP domain verified by ChIP. The functionally unknown SCOP domain of Rtt103 might have the several roles in transcription termination via Rat1.
