Abstract
Cyclopeptenone prostaglandin은 항 염증작용과 세포 보호작용을 포함한 여러 가지 생리학적 기능을 가지고 있다. 15-deoxy-△12,14-prostaglandin J2 (15d-PGJ2)는 α,β-unsaturated carbonyl moiety를 가지고 있는 대표적인 cyclopeptenone prostaglandin의 하나로, 세포의 분열과 사멸에 있어서 이중적인 효과를 나타내는 것으로 알려져 있다. 또한 본 실험의 선행연구를 통해 유방암 조직에서의 15d-PGJ2의 축적농도가 정상조직에 비해 높은 수준으로 존재한다는 것을 밝혔다. 본 연구에서는 인체 유방암 세포 (MCF-7) 에서 15d-PGJ2가 세포 성장 촉진인자로 작용하는 단백질인 Akt를 활성화시키는 분자 기전을 연구하고자 하였다. MCF-7 세포에 15d-PGJ2를 처리하였을 때 Akt의 인산화가 유도되는 것을 확인하였다. 한편, Akt의 활성은 종양 억제 인자인 phosphatase and tensin homolog (PTEN)에 의해 저해된다. MCF-7 세포에서 PTEN의 유전자에 대한 siRNA를 이용해 단백질의 합성을 억제시켰을 때, Akt의 인산화가 현저히 증가하는 것을 관찰하였다. 15d-PGJ2는 구조적으로 활성이 높은 α,β-unsaturated carbonyl group을 가지고 있어 다양한 단백질의 cysteine thiol group과 Michael reaction을 통해 결합할 수 있다. 이를 바탕으로 하여, 본 연구에서는 15d-PGJ2에 의해 유도되는 Akt의 인산화는 PTEN과 15d-PGJ2와의 상호작용을 통한 PTEN의 구조적 변형에 의한 것으로 가정하고 실험을 진행하였다. Biotin이 결합된 15d-PGJ2를 MCF-7 세포에 처리하여 내재성 PTEN과의 결합을 확인하였다. PTEN의 cysteine 124는 효소의 활성부위에 존재하며, 산화-환원 조절에서 중요하게 작용하는 잔기로 알려져 있다. MCF-7 세포에 PTEN의 cysteine 124가 공유변이에 중요한 역할을 하는지 알아보기 위해 PTEN의 cysteine 124를 serine으로 치환한 벡터(C124S-PTEN)를 이용하여 결합여부를 알아보았다. 15d-PGJ2와 PTEN의 결합은 C124S-PTEN 돌연변이를 주입시킨 세포에서 현저히 감소함을 관찰하였다. Docking study또한 15d-PGJ2가 반응하는 부위로서 PTEN의 cysteine 124가 중요함을 뒷받침하였다. 결론적으로 15d-PGJ2는 PTEN의 cysteine 124와 결합을 하여 MCF-7세포에서 Akt 의 인산화를 유도하며, 그로 인해 암화 과정이 촉진될 것으로 사료된다.
Cyclopentenone prostaglandins (cyPGs) have diverse physiologocal functions including anti-inflammatory and cytoprotective effects. 15-Deoxy-12,14-prostaglandin J2 (15d-PGJ2), a representative J-series cyPG that contains an α,β-unsaturated carbonyl moiety, has bi-phasic roles in cell proliferation and apoptosis. In the present study, I have investigated the role of 15d-PGJ2 in the activation of Akt, a cell survival protein, and elucidated its underlying molecular mechanisms. Treatment of human breast cancer (MCF-7) cells with 15d-PGJ2 led to induction of Akt phosphorylation. Akt is negatively regulated by phosphatase and tensin homolog (PTEN). The si-RNA-mediated silencing of PTEN caused the elevation of Akt phosphorylation in MCF-7 cells. Since the α,β-unsaturated carbonyl moiety in the cyclopentenone ring of 15d-PGJ2 is known to cause thiol modification of various proteins, I hypothesize that 15d-PGJ2-induced Akt phosphorylation may result from the covalent modification of PTEN by this cyPG. Covalent binding of 15d-PGJ2 to PTEN was verified by incubation of endogenous PTEN with biotin-conjugated 15d-PGJ2. PTEN harbors cysteine 124 (Cys124) in its catalytic domain, which is subjected to redox modulation. To determine whether Cys124 is a critical site for covalent modification by 15d-PGJ2, MCF7 cells were transiently transfected with expression vectors for wild-type PTEN or mutants PTEN in which cysteine were replaced by serine. PTEN binding after treatment with biotinylated 15d-PGJ2 was abolished in the mutant cells (C124S-PTEN). A docking study further supports that Cys124 of PTEN is a putative target of 15d-PGJ2. These observations suggest that 15d-PGJ2 binds to PTEN presumably at Cys124, thereby stimulating Akt phosphorylation in MCF-7 cells, which may account for the oncogenic role of 15d-PGJ2 in human breast carcinogenesis.
