Abstract
To improve stem cell survival rate, NS3694 delivery systems were developed using liposomes as carriers. NS3694, an apoptosome inhibitor, was used for enhancing the stem cell survival rates. The sizes of NS3694-entrapped liposomes were measured by light scattering methods. Liposomal encapsulation efficiey of NS3694 was 70%. The viability of adipocyte-derived mesenchymal stem cells was observed to be significantly higher following treatment with liposomal NS3694 as compared to the treatment with empty liposomes. The treatment of liposomal NS3694 did not affect the differentiation of stem cells over 7 days. In mice, following in vivo injection of adipocyte-derived stem cells, the co-treatment with liposomal NS3694 showed significantly higher DNA contents than other groups. Moreover, in rabbits, the in vivo injection of adipocyte-derived stem cells with liposomal NS3694 exhibited higher cellular DNA contents than the group treated with free NS3694. Taken together, these results suggest that the liposomal NS3694 might be used for the in vivo stem cell delivery by increasing stem cell viability.
