Abstract
Enterococcus faecalis는 구강 내 상주균이면서 동시에 난치성 치근단 치주염(refractory apical periodontitis)의 주 원인균이다. 리포테이코익산(lipoteichoic acid; LTA)은 E. faecalis의 중요한 병독인자로서 구강에서 E. faecalis LTA(Ef.LTA)에 대한 인지는 다양한 숙주 반응을 위한 필수적인 첫 단계이다. 구강을 덮고 있는 타액 내의 단백질들은 세균의 병인기전 및 인체의 면역반응 초기에 중요한 역할을 할 것으로 예측되며, 이에 따라 Ef.LTA와 특이적으로 부착하는 타액 내 리포테이코익산-부착 단백질(Ef.LTA-Binding proteins)을 동정하여 E. faecalis의 병인기전을 규명하기 위한 기반지식을 제공하고자 한다.
연구에 사용한 Ef.LTA는 streptavidin-FITC나 neutravidin agarose bead와 특이적 결합이 가능토록 biotin을 부착시켰으며, biotin의 정상적 결합여부는 Toll유사수용체 2가 발현된 CHO 세포주를 이용한 binding assay를 통하여 확인하였으며, biotin이 부착된 Ef.LTA의 염증유발능력을 마우스 대식세포주인 RAW 264.7 세포주에 처리한 후 염증관련인자의 발현을 통해 확인하였다. 채취한 사람 타액을 Ef.LTA-biotin과 혼합하여 Ef.LTA-부착 단백질이 Ef.LTA-biotin에 부착하도록 한 후 neutravidin agarose bead를 이용하여
타액중의 Ef.LTA-BPs을 분리하였고 이를 푸리에 변환 이온트랩 사중극자 질량분석(LTQ-Orbitrap hybrid fourier transform mass spectroscopy;LTQ-FT/MS)을 이용하여 동정하였다. 총 68종류의 단백질이 확인되었으며, 그중 통계적 유의성이 있는 11종류의 Ef.LTA-BPs이 밝혀졌으며 이중 Lysozyme C, Apolipoprotein A-I, Lipocalin-1, Short palate, lung and nasal epithelium carcinoma-associated protein 2, Bactericidal/permeability increasing protein-like 1, Zymogen granule protein 16 homolog B, Hemoglobin subunit α, Hemoglobin subunit β은 타액에 정상적으로 존재하는 단백질로서 항균이나 항염증작용 또는 병원체 인지나 염증반응촉진 등 다양한 역할을 하는 것으로 알려져있다. 이러한 Ef.LTA-BPs의 확인은, E. faecalis의 병인기전을 이해하고 나아가 LTA에 의한 질병의 진단, 예방, 치료 방법을 계발하는데 도움이 될 것으로 생각한다.
Enterococcus faecalis (E. faecalis ) is an oral commensal bacterium and closely related with refractory apical periodontitis. Lipoteichoic acid (LTA) is one of the major virulence factor of E. faecalis , it plays important roles in bacteiral adhesion, biofilm formation and its pathogenesis to induce inflammation leading to tissue damage in the host. Although the oral cavity is the initial site to recognize E. faecalis LTA(Ef.LTA), the salivary proteins related with pathogenesis of Ef.LTA are poorly understood. In this study, we isolate the salivary proteins with affinity to Ef.LTA and identified them using a high-resolution mass spectrometry. The purified Ef.LTA was biotinylated using Biotin-3-sulfo-N-hydroxysuccinimid ester sodium salt (Ef.LTA-biotin), and the conjugation of Ef.LTA with biotin were confirmed using binding assay on CHO/CD14/TLR2 cells. To examine whether Ef.LTA-biotin still retains its immunostimulating activity as Ef.LTA, RAW 264.7 cells were stimulated with Ef.LTA-biotin or Ef.LTA for 24h and the production of nitric oxide (NO) and MIP-1α were determined. In the result, Ef.LTA-biotin but not Ef.LTA were detected on CHO/CD14/TLR2 cell after streptavidin-FITC treatment and Ef.LTA-biotin could induce the production of NO and MIP-1 α in RAW 264.7 cells. To capture and identify the Ef.LTA-binding proteins (Ef.LTA-BPs) in saliva, 900 ug of
saliva from 9 healthy subjects were incubated with Ef.LTA-biotin for 1 h and the Ef.LTA-BPs were isolated using neutravidin-beads (NAbeads). The isolated Ef.LTA-BPs were visualized on SDS-PAGE gel and were identified using LTQ-Orbitrap hybrid fourier transform (FT) mass spectroscopy (MS). Eleven proteins of Ef.LTA-BPs with statistical significance (p<0.05) were identified including Lysozyme C, Apolipoprotein A-I, Lipocalin-1, Short palate, lung and nasal epithelium carcinoma-associated protein 2, Bactericidal/permeability increasing protein-like 1, Zymogen granule protein 16 homolog B, Hemoglobin subunit α and Hemoglobin subunit β. These Ef.LTA-BPs could be categorized into tree in relation to 1) anti-microbial function 2)antiimflammation function, and 3) recognition of pathogen or stimulation of inflammation.
