코발트전극과 비색법을 이용한 양액 내 인산 이온 센싱 기술 개발 : Development of Hydroponic Phosphate Sensing Technology using Cobalt Electrodes and Colorimetric Method

Abstract
인산은 작물의 성장에 있어 생장점의 중요한 요소 중 하나로, 수정과 개화에 직접적인 영향을 미치기 때문에 양액 내 적합한 인산 이온 농도를 유지하는 것이 중요하다. 현재 사용되고 있는 시스템은 전체 이온의 전기전도도(EC)를 측정하는데, 이는 전체 무기이온들의 총 합을 예측 하는 방법으로 모든 이온 성분을 비례적으로 증감시키기 때문에 작물의 성장에 따라 변하는 각 이온의 흡수 변화를 개별적으로 관리하기가 어렵다. 특히, 인산은 용액 내 pH 에 따라 다양한 이온 형태를 갖고, 인산 이온에 선택적으로 반응하는 감지 물질의 상용화가 이루어지지 않아 양액에 적용할 수 있는 인산 이온 측정 기술의 개발이 필요하다. 선행 연구에서 수행된 코발트 전극을 이용한 인산 측정은 넓은 농도 대역에서는 측정 가능성을 보였지만, 50 mg/L 내외의 저농도에서는 전극의 기전력 안정성이 떨어져 측정 성능이 저하되는 문제점이 나타났다. 인산을 측정하는 또 다른 방법으로, 인산에 대한 발색체 형성을 이용한 몰리브덴 비색법이 있는데, 저농도의 인산 이온 측정이 가능하지만, 상대적으로 고농도 구간에서는 감도가 낮은 문제가 존재한다. 본 연구에서는 실제 양액 측정에 활용할 수 있는 인산 이온 측정 기술을 개발하였으며, 몰리브덴 비색법과 코발트 전극을 각각 이용하여 측정 성능을 확인하였으며, 두 가지 방법을 합친 다중선형회귀 캘리브레이션 모델식을 개발하여 미지 양액 시료의 인산 이온 농도 측정 예측 성능을 검증하였다. 현재 사용되고 있는 양액과 배액의 농도를 고려하여 Hoagland 양액 조성을 기반으로 인산 이온에 대해 0 – 200 mg/L 범위를 갖는 시료들을 제작하여 몰리브덴 비색법을 적용하였을 때 스펙트럼 분석기를 이용하여 450 – 1100 nm의 파장대에서 양액 내 인산 이온 농도와 상관관계가 높은 파장 대역을 선별하였다. 선발된 파장을 기반으로 수립된 회귀식을 통해 미지의 양액 시료 농도를 예측하였을 때, 표준 분석실 분석 값 대비 예측 값 관계식은 결정계수가 0.89의 높은 선형성을 나타내었고 평균제곱근 오차는 20 mg/L를 보였다. 동일한 시료들을 기반으로 수행한 코발트 전극 기반 인산 농도 예측에서는2점정규화를 통해 전극의 신호 드리프트를 보정하여 결정계수(R2) 0.93와 평균제곱근 오차(RMSE)는 13.0 mg/L으로 나타났다. 이에 두 방법의 장점을 갖춘 측정 방법으로 다중선형회귀인 MLR 기반으로 개발한 퓨전 모델을 적용하였을 때, 다중선형회귀 모델은 결정계수(R2) 0.96와 평균제곱근 오차(RMSE)는 10.17 mg/L 로 기존 측정방법 대비 개선된 성능을 나타내었다. 향후 퓨전 방식 기반의 현장형 인산 측정 시스템을 개발하여 양액 측정에 활용할 경우 양액 내 인산 관리를 보다 정밀하게 수행하여 작물 생육 및 생산성 증진에 기여할 수 있을 것으로 기대한다.
It is important to maintain the proper concentration of phosphate ion in hydroponic nutrient solutions because phosphate ion is one of the important growth factors for crop growth, which is directly relating to pollination and flowering. Current hydroponic systems usually measure electrical conductivity (EC) of nutrient solutions, which is related to the total sum of ions in nutrient solutions. However, EC proportionally decreases or increases according to all ionic components, thereby limiting the precise management of nutrient ions that are differently affected by the crop growth. Particularly, it is needed to develop an accurate and precise phosphate ion measurement technology that can be applied to the nutrient solution because phosphate has a variety of ionic forms depending on the pH in the solution and there is no commercial sensing material that can selectively react with phosphate ions in nutrient solutions.
In previous studies, phosphate measurement using the cobalt electrode showed the possibility of measurement in a wide concentration range. However, the EMF of the electrode was unstable at the concentration of about 50 mg/L, resulting in poor measurement performance. Colorimetric method using molybdenum measured the low concentration of phosphate ions, but showed a low sensitivity in a relatively high concentration range. In this study, we developed a phosphate ion sensing technology that can be used to measure the phosphate ions in actual nutrient solutions. The measurement performances of the colorimetric method and cobalt electrode method were evaluated respectively, then a fusion model based on the two methods was developed using a multi linear regression (MLR). The predictability of the fusion model for phosphate ion concentrations in nutrient solutions, several unknown nutrient solutions were sampled and measured. Specifically, the colorimetric method was applied to the samples with the range of 0 – 200 mg/L of phosphate ions, which were prepared based on the Hoaglands composition. Then, an optimal wavelength for predicting the concentration of phosphate ions in nutrient solutions was investigated using a spectrometer with the spectrum range of VIS-NIR (450 – 1100 nm). After the determination of the optimal wavelength, the predicted phosphate concentrations based on the colorimetric method showed a linear relationship with a coefficient of determination (R2) of 0.89 and a root mean square error (RMSE) of 20 mg/L for the actual phosphate concentrations of nutrient solution samples. In case of the cobalt electrode based measurements with the two-point normalization, it showed a R2 of 0.93 and a RMSE of 13.0 mg/L. Compared to the conventional methods, the fusion model showed the improved predictability with a R2 of 0.96 and a RMSE of 10.17 mg/L. The results showed the phosphate ion management in nutrient solutions can be performed more precisely by developing an on-site measurement system with the fusion model. Through the precise management of the phosphate ions in nutrient solutions, it would be possible to improve crop growth and productivity.
