Abstract
최근 실크 피브로인 하이드로젤을 제작함에 있어 horseradish peroxidase(HRP)와 과산화수소에 의한 효소 가교 방법이 주목 받고 있다. HRP에 의한 가교로 하이드로젤을 제작하면 탄성이 있으며 투명한 하이드로젤을 제작할 수 있으나, 과량의 과산화수소가 존재하면 HRP의 비활성화가 일어난다. 따라서 본 연구에서는 직접적인 과산화수소의 부가를 피하기 위해 연속 효소 반응을 적용하여 포도당과 glucose oxidase(GOx)에 의해 실크 피브로인의 젤화를 개시하였다. 연속 효소 반응에서는 먼저 GOx가 포도당과 산소를 각각 글루코노-델타-락톤과 과산화수소로 전환시키고, 이 반응에서 생성된 과산화수소를 기질로 하여 HRP가 실크 피브로인 내의 타이로신 잔기를 산화하여 다이-타이로신 반응을 촉진한다.
연속 효소 가교 하이드로젤(SF/GOx/HRP)의 특성 분석은 크게 두 가지로 나누어, 초기 젤화 거동과 제작한 하이드로젤의 역학적 성질에 대해 살펴보았다. SF/GOx/HRP의 젤화 시간은 SF/HRP에 비해 약 2.7 배 단축되었는데, 이에 대한 자세한 분석을 위해 반응계 내에 존재하는 화합물들의 영향을 자세히 알아보았다. 그 결과, 과산화수소의 간접적인 공급으로 기존에 제기되었던 HRP의 비활성화 문제를 피할 수 있었던 것으로 생각된다. 한편, SF/GOx/HRP의 역학적 성질을 측정하였을 때 초반에는 탄성률이 낮았으나 시간이 경과함에 따라 베타 시트 구조가 발달하며 탄성률이 증가하였다. 하지만 최종적인 탄성률은 SF/HRP에 비해 낮았으며 이는 내부 구조가 비교적 조밀하게 형성되지 않았기 때문이었다. SF/GOx/HRP는 포도당이 존재할 경우에 GOx에 의해 과산화수소가 방출되는 특징을 가지고 있어 항균성이 나타났다. 이에 따라 향후 지혈제로서 활용될 수 있을 것으로 기대된다.
The enzymatic crosslinking reaction by horseradish peroxidase(HRP) and hydrogen peroxide(H2O2) is getting attention for fabricating silk fibroin hydrogel. HRP-mediated crosslinked hydrogel is elastic and optically transparent, yet has the drawback of HRP inactivation by excess H2O2. In this study, the tandem enzyme reaction was applied to avoid the direct addition of H2O2, which triggered the gelation of silk fibroin by glucose and glucose oxidase(GOx). In the tandem enzyme reaction with GOx and HRP, GOx converts glucose and oxygen into glucono-δ-lactone and H2O2, and subsequently, HRP consumes indirectly provided H2O2. In consequence, HRP oxidizes tyrosine residue and induces di-tyrosine linkage of silk fibroin.
The properties of serial enzymatic crosslinked hydrogels(SF/GOx/HRP) were analyzed by dividing them into two parts: initial gelation behavior and time-dependent stiffening. The effects of chemicals in the reaction system were investigated since the gelation time of SF/GOx/HRP was approximately 2.7 times shorter than SF/HRP. To sum up, the indirect supply of H2O2 can inhibit HRP deactivation. The elasticity was increased over time despite lower modulus than SF/HRP at 0 hr because of the β-sheet development. However, the final modulus of SF/GOx/HRP was lower than that of SF/HRP because the pore structure was not relatively dense. In addition, SF/GOx/HRP showed antibacterial activity because hydrogel releases H2O2 by GOx when glucose was supplied. As a result, it is expected to be used for glucose sensing hydrogel or hemostatic agent.
