Abstract
세포 내 DNA 이중가닥 절단은 독성이 매우 크기 때문에 신속하고 정확하게 수선되어야 한다. RecQ helicase family 중 하나인 RecQL4는 유전체 안정성에 중요하며, 변이가 일어나면 Rothmund-Thomson Syndrome(RTS)등의 유전질환이 발생한다. 선행연구 결과, RecQL4는 이중가닥 절단 반응에서 핵심적인 역할을 담당하는 Mre11-Rad50-Nbs1(MRN) 복합체의 안정성 유지에 필요하며, RecQL4의 MRN 복합체 안정성 유지 작용에는 탈유비퀴틴화효소인 USP28이 관여하는 것으로 확인되었다. 그러나 정상적인 세포에서 USP28의 기능이 억제되어도 MRN 복합체의 안정성에는 아무런 변화가 없었으며, 이는 새로운 탈유비퀴틴화효소의 존재를 시사한다.
본 연구에서는 MRN 복합체의 안정성에 기여하는 새로운 탈유비퀴틴화효소를 선별하기 위해 현재까지 알려진 50여 종의 탈유비퀴틴화효소 중 DNA 손상, 세포주기, 발암과 관련된 10개의 탈유비퀴틴화효소를 후보 단백질로 선정하여 RecQL4의 기능이 억제된 세포에 형질주입한 후 면역염색법으로 MRN 복합체의 안정성을 확인하였다. 그 결과, 기존에 연구되었던 USP28을 형질주입한 것과 같이 USP2를 형질주입한 세포에서 MRN 복합체의 안정성이 증가하는 것을 확인하였다.
또한, USP2를 과발현시킨 세포에서는 RecQL4에 결함이 있는 세포에서 관찰되었던 ATM 활성화와 상동재조합 수선의 결함도 회복됨을 확인하였다. 특히, RecQL4가 발현되는 정상세포에서 USP2와 USP28의 기능을 각각 억제하였을 때는 ATM 활성화와 상동재조합 수선이 영향을 받지 않았지만, USP2와 USP28의 기능을 동시에 억제하면 MRN 복합체의 불안정성이 증가하고 ATM 활성화와 상동재조합 수선이 진행되지 않음을 관찰하였다. 이는 이 두 탈유비퀴틴화 효소가 RecQL4 의존적으로 MRN 복합체의 안정성 유지에 기여하는 중복된 기능을 수행함을 의미한다. 또한, USP2의 이중가닥 절단 부위 결합 기전을 조사한 결과, USP2가 RecQL4 의존적으로 이중가닥 절단 부위에 결합하며, 그 결합에는 ATM kinase의 활성이 필요함을 확인하였다.
본 연구는 DNA 이중가닥 절단에 대한 세포반응에서 RecQL4 의존적으로 MRN 복합체의 안정성 유지에 기여하는 새로운 탈유비퀴틴화효소를 찾아냈으며, RecQL4의 세포 내 기능 규명에 기여하여 RTS 환자의 증상을 이해하고 치료법을 제시할 수 있는 단서를 제공했다는 점에서 큰 의미가 있다.
Intracellular DNA double strand breaks are highly toxic and must be repaired quickly and accurately. RecQL4, one of the RecQ helicase family, is important for genome stability. The mutations in RecQL4 occurs, a genetic disorder called Rothmund-Thomson Syndrome (RTS). According to the previous studies, RecQL4 is required to maintain the stability of the Mre11-Rad50-Nbs1 (MRN) complex, which plays a key role in DNA double strand breaks repair. Moreover, it was confirmed that USP28, a deubiquitination enzyme, is involved in maintenance of stability of the MRN complex of RecQL4. However, when the function of USP28 was inhibited in normal cells, there was no change in the stability of the MRN complex, suggesting the presence of a new other involved deubiquitinase.
In this study, in order to find new deubiquitinases that contribute to the stability of the MRN complex, 10 deubiquitinases related to DNA damage, DNA repair, and cell cycle among 50 deubiquitinases which known to date were selected as candidate proteins. Then, after the deubiquitinases were transfected into cells with RecQL4 function suppressed, the stability of the MRN complex was confirmed by immunostaining. As a result, it was confirmed that the stability of the MRN complex increased in cells transfected with USP2 as in the case of transfection with USP28, which was previously confirmed.
In addition, it was found that the cells overexpressing USP2 recovered the defects of ATM activation and homologous recombination observed in cells with RecQL4 defects. In particular, in cells that inhibited the functions of USP2 and USP28, respectively, in normal cells expressing RecQL4, ATM activation and homologous recombination repair were not affected. However, it was observed that in cells with the functions of USP2 and USP28 simultaneously inhibited, not only the instability of the MRN complex increased but also ATM activation and Homologous recombination repair did not proceed properly. This means that these two deubiquitinases perform a redundant function of maintaining the stability of the MRN complex dependent on RecQL4. In addition, as a result of examining the mechanism of binding of the double strand breaks site of USP2, it was confirmed that USP2 binds to the double strand breaks site dependent on RecQL4, and that binding requires the activity of ATM kinase.
This study found a new deubiquitinase that contributes to the stability of the MRN complex dependent on RecQL4 in the cellular response to DNA double strand breaks. In addition, it has a great significance in that this study has provided clues to understand the symptoms of RTS syndrome and suggest treatments by contributing to the investigation of the intracellular function of RecQL4.
