- Abstract
- This study attempt to have improved cell membrane permeability with higher biocompatibility of diLR10 peptides (CPPs). As a remedy, fatty acid bonded form, monomer Cx-LR10 (x= number of carbon) was synthesized which expected to increase the half-life in the blood. When measuring the cell penetration efficiency by flow cytometry analysis, the lipidated dimer bundle had superior cellular permeability compared to the conventional non-lipidated diLR10. And, we anticipated that this result would be related to the formation of oligomer. We monitored that the tendencies of oligomer formation by length of fatty acid were consistent with the tendencies of cell permeability to vary with the length of fatty acids. Therefore, increasing the hydrophilicity of peptides enhanced the degree of oligomer formation, which can also affect cell penetration. Taken together, lipidated dimer bundle peptide, acCx-LR10, showed improved cell permeability in eukaryotic cells and this effect can be interpreted by oligomer formation study. It suggest that this form of lipidated dimer bundle is essential for the results to appear.
본 연구는 사전 연구를 통해 발견한 낮은 농도의 빠른 시간에 세포막 투과가 가능하고 낮은 독성의 diLR10 펩타이드 (CPP)의 높은 생체 적합성을 갖추기 위한 시도를 하고자 한다. 혈액 내 머무름 시간(half-life)을 늘리기 위하여 지방산을 결합시키는 리피데이션을 시켰고 이를 시퀀스 내 시스테인(Cys)간 이황화결합을 시켜 이량체(다이머 번들)로 만들 수 있었다. 리피데이션 된 헤테로 다이머가 기존의 diLR10에 비해 세포투과성이 우수하다는 점을 발견하였고 이어진 올리고머 형성도 실험을 통해 올리고머의 형성이 세포투과성에 직접적인 영향을 주는 것을 확인하였다. 이러한 올리고머 형성은 펩타이드의 소수성에 의해 활성화 되거나 또는 억제되는 것 같이 나타났는데, 소수성을 지방산의 길이로 조절하여 가장 올리고머 형성을 극대화 시킬 수 있는 지방산의 길이를 찾았다. 결과적으로 적정 지방산의 길이를 사용하여 리피데이션 된 다이머 형태의 세포 안으로 들어갈 수 있는 효과가 향상된 전달물질을 개발하였다.
