Abstract
physiological characteristics of the existing sugars. Maltogenic amylase from
Bacillus stearothermophilus (BSMA) catalyzes the transglycosylation reaction in
the presence of suitable glycosyl acceptors.
In this study, production of maltosyl-trehalose by transglycosylation reaction,
structural determination of transferred product, and characterization of
physicochemical properties were carried out.
Maltosyl-trehalose was produced by maltogenic amylase from Bacillus
stearothermphilus through the reaction with maltotriose as the maltosyl donor
and trehalose as its acceptor. It was identified by thin layer chromatography
and high performance ion chromatography. The transfer product was purified with
Bio-Gel P-2 column chromatography and recycled prep high pressure liquid
chromatography. The structure was identified by 13C-NMR analysis, matrixassisted
laser desorption ionization with time-of-flight/mass spectrometry and
liquid chromatography-mass spectrometry.
Chemical shift of maltosyl-trehalose in 13C-NMR spectrum was compared with that
of the trehalose. The chemical shift of one of two C-6 in trehalose changed from
61.39 to 44.48 and 66.78 for maltosyl-trehalose. The molecular mass was
estimated to be 666 daltons from the MALDI-TOF analysis. These results support
that the transglycosylated product was maltosyl-trehalose, in which maltose was
attached by an ¥á-(1, 6) glycosidic linkage to trehalose. Optical rotation
([¥á]D) of maltosyl-trehalose was +155.7¢ª at 20¡É. Flow behavior of 10, 20, and
30% solution of maltosyl-trehalose showed typical Newtonian fluid
characteristics. Sorption isotherm of maltosyl-trehalose displayed a typical
type¥² isotherm curve. Glass-transition temperatures of maximally freezeconcentrated
solutes (T''g), and T''m were observed at -49 and -42¡É,
respectively. It also exhibited medium hygroscopicity and very low Maillard
reactivity.
