Abstract
비만은 Th1 및 Th17 세포 수 증가를 특징으로 하는 T 세포의 분화 이상을 일으키며, 비타민 D 대사의 조절 이상과도 관련이 있는 것으로 알려져 있다. 비만마우스의 지방 조직 및 비장 세포에서 Th1, Th17 세포의 수가 증가하는데, 두 세포에서 특징적으로 분비되는 IFN-γ와 IL-17는 자가 면역 질환의 발병 및 유지와 밀접한 관련이 있는 것으로 보고되고 있다. 한편, 비타민 D는 Th1 세포 및 Th17 세포로의 분화와 사이토카인 생성을 억제한다. 비만과 비타민 D 모두 자가포식에 영향을 미치는 것으로 보고되었으나, 이는 주로 면역세포 외에 췌장, 간, 심장 및 지방 조직에 관한 연구이며, 비만마우스의 T 세포 내 자가포식에 대한 비타민 D의 효과를 확인한 연구는 부족한 실정이다. 따라서, 본 연구에서는 비타민 D의 in vitro 처리가 고지방식이로 유도한 비만마우스의 T 세포의 증식, 기능 및 자가포식에 미치는 영향을 확인하는 것을 목적으로 하였다. 5주령 수컷 C57BL/6 마우스를 두 군으로 임의 배정한 후 대조식이 또는 고지방식이 (10% 또는 45% kcal fat, CON 또는 HFD)를 12주 동안 공급하였다. 희생 후 비장에서 negative selection 방법으로 T 세포를 분리하여, 72시간 동안 in vitro 상에서 활성형 비타민 D인 1,25-dihydroxyvitamin D3 [1,25(OH)2D3]의 최종 농도가 10 nM이 되도록 처리하였다. 대조 배지에는 1,25(OH)2D3 용해 시 사용된 ethanol을 동량 추가하였다. TCR 자극을 위해 배양 기간 동안 anti-CD3 mAb, anti-CD28 mAb를 처리하였다. T 세포의 증식 정도는 [3H]TdR가 DNA로 혼입된 양을 정량화하였다. 배양 후 회수된 T 세포에서 유세포분석을 통해 CD25, RORγt 및 Foxp3의 표현형과 세포 내 발현량을 확인하였고, Western blot 분석으로 자가포식 관련 단백질의 발현을 측정하였다. 또한, CD4 T 세포 아형에서 특징적으로 분비되는 사이토카인인 IFN-γ, IL-17A, IL-4, IL-10의 생성량을 분석하였다. HFD군에서 T 세포의 증식이 낮은 경향이 있었으나, IFN-γ의 생성량과 LC3 II/I의 비율은 CON군에 비해 높았다. 비만마우스의 T 세포에서 자가포식 전구체의 가용성이 감소하여 자가포식이 억제된 것으로 사료되며, 높은 수준의 Th1 사이토카인 생성에 의해 LC3의 발현이 상향 조절 되었을 가능성이 있다. 한편, 대조마우스군 뿐만 아니라 비만마우스군에서도 1,25(OH)2D3 처리에 의해 IL-17A 분비량이 유의하게 억제된 결과를 통해, 질병 상태에서 비만에 의해 야기되는 Th17 세포로의 과도한 분화와 기능 이상이 비타민 D 보충으로 조절될 가능성이 있음을 확인하였다. 본 연구의 결과는 식이로 유도된 비만이 T 세포의 기능에 영향을 미치고 자가포식을 억제하여 T 세포 항상성의 조절 장애를 유발할 수 있음을 시사하며, 이는 비만에서 주로 관찰되는 염증에 기여하는 요인일 가능성을 시사한다.
Obesity is associated with impaired regulation of T cells characterized by increased number of Th1 and Th17 cells and dysregulation of vitamin D metabolism. Both obesity and vitamin D has been reported to affect the autophagy, however, study which investigated the effects of vitamin D on autophagy in T cells from obese mice is limited. Therefore, we determined whether in vitro treatment with vitamin D affects proliferation, function, and autophagy of T cells from obese and control mice. Five-week-old male C57BL/6 mice were fed control or high-fat diet (10% or 45% kcal fat: CON or HFD) for 12 weeks. T cells purified by negative selection from splenocytes were stimulated with anti-CD3 and anti-CD28 mAbs and cultured with either 10 nM 1,25(OH)2D3 or vehicle control. Proliferative response, expression of CD25, Foxp3, RORγt and autophagy-related proteins (LC3A/B, SQSTM1/P62, BECLIN-1, ATG12) in T cells and production of IFN-γ, IL-4, IL-17A and IL-10 by T cells were measured. In HFD group, the proliferation of T cells tended to be lower, but the IFN-γ production level and the ratio of LC3 II/I were higher compared with CON group. The elevated Th1 cytokine production in HFD group may have contributed to the increased conversion of LC3-I to LC3-II. However, effect of vitamin D treatment on autophagy of T cells was not observed. As 1,25(OH)2D3 significantly inhibited the secretion of IL-17A in both CON group and HFD groups, we confirmed the possibility that abnormal regulation of T cell function caused by obesity in the inflammatory state may improve by vitamin D supplementation. Our findings suggest that diet-induced obesity may impairs function and inhibits autophagy of T cells, possibly leading to dysregulation of T cell function, which may be the factor aggravating inflammation commonly observed with obesity.
